[Search for Neuroprotective Compounds -From 4-Methycatechol to Citrus Compounds]
1Department of Pharmaceutical Pharmacology, College of Pharmaceutical Sciences, Matsuyama University.
Citrus peel compounds, heptamethoxyflavone and auraptene, show neuroprotective effects by reducing inflammation and inducing neurotrophic factors. Kawachi Bankan juice, rich in auraptene, may help suppress cognitive dysfunction in aging individuals.
Area of Science:
- Neuroscience
- Pharmacology
- Biochemistry
Background:
- Development of an enzyme immunoassay (EIA) system for mouse nerve growth factor (NGF) in the 1980s facilitated research into NGF's physiological roles and synthesis regulation.
- Low molecular weight compounds, like 4-methylcatechol, were identified for their ability to stimulate NGF synthesis, showing therapeutic potential for neuronal degeneration in the central and peripheral nervous systems.
Purpose of the Study:
- To identify and isolate neuroprotective components from citrus peels.
- To evaluate the neuroprotective potential of specific compounds, 3,5,6,7,8,3',4'-heptamethoxyflavone (HMF) and auraptene (AUR), in the central nervous system (CNS).
- To assess the efficacy of Kawachi Bankan juice, enriched with AUR, in preventing cognitive dysfunction.
Main Methods:
- Isolation and identification of neuroprotective compounds from citrus peels.
- In vitro and in vivo studies using pathological model mice to evaluate neuroprotective effects.
- Analysis of compound content in citrus peels and juice.
- Clinical assessment of cognitive function in aged volunteers.
Main Results:
- 3,5,6,7,8,3',4'-heptamethoxyflavone (HMF) demonstrated neuroprotective ability in the CNS by inducing brain-derived neurotrophic factor (BDNF) and suppressing inflammation.
- Auraptene (AUR) also exhibited CNS neuroprotective effects through inflammation suppression and potential induction of neurotrophic factors.
- Kawachi Bankan peel powder showed neuroprotective effects in pathological model mice; AUR was found to transfer from peel to juice during processing. Enriched Kawachi Bankan juice suppressed cognitive dysfunction in aged volunteers.
Conclusions:
- Citrus peel compounds HMF and AUR possess significant neuroprotective properties beneficial for CNS health.
- Auraptene-enriched Kawachi Bankan juice presents a practical, natural approach for mitigating age-related cognitive decline.
