Summary

Interferon-alpha (IFN-alpha) treatment increases parallel tubular structures (PTS) in specific immune cells. These IFN-alpha-induced changes in OKT 8+ cells correlate with enhanced natural killer cell activity.

Background:

  • Interferon-alpha (IFN-alpha) is known to modulate immune cell function.
  • Natural killer (NK) cell activity, particularly of large granular lymphocytes (LGLs), is reportedly augmented by IFN.
  • Some LGLs contain parallel tubular structures (PTS) and/or electron-dense granules.

Purpose of the Study:

  • To investigate the induction of parallel tubular structures (PTS) and/or electron-dense granules in peripheral blood lymphocytes by IFN-alpha.
  • To determine the relationship between IFN-alpha, PTS, and specific cell surface antigens (OKT 8, Leu-7) on lymphocytes.
  • To assess the impact of IFN-alpha on lymphocyte cytotoxicity.

Main Methods:

  • Incubation of normal peripheral blood mononuclear cells with IFN-alpha.

  • Main Results:

    • IFN-alpha treatment did not alter surface antigen reactivity with OKT 3, 4, or 8, and Anti-Leu-7 (HNK-1) antibodies.
    • Electron microscopy revealed a significant increase in PTS-positive cells among OKT 8+ and Anti-Leu-7+ populations after IFN-alpha exposure.
    • Cytotoxicity assays demonstrated enhanced lytic activity against the K562 cell line.

    Conclusions:

    • IFN-alpha primarily induces PTS and/or associated structures in OKT 8+ lymphocytes.
    • Cells coexpressing OKT 8 and Leu-7 antigens may contribute to a minor increase in PTS.
    • The observed increase in PTS+/OKT 8+ cells likely contributes to the enhanced cell cytotoxicity induced by IFN-alpha.
