Drug Delivery: Miscellaneous Routes01:22

Drug Delivery: Miscellaneous Routes

601
Drug delivery methods like oral inhalation, nasal sprays, transdermal patches, eye drops, intravitreal injection,  and rectal administration provide localized effects with reduced toxicity.
Oral inhalation and nasal sprays swiftly transfer drugs across the respiratory epithelium's mucosal layer. Inhaled glucocorticoids and bronchodilators directly target lung conditions such as asthma, while fluticasone nasal spray mitigates allergic rhinitis.

Antiasthma Drugs: Mast Cell Stabilizers and Anti-IgE Drugs

1.5K
Asthma is a chronic respiratory condition for which new therapeutic avenues, including anti-inflammatory drugs like mast cell stabilizers and anti-IgE treatments, continue to be developed.
Mast cell stabilizers, such as cromolyn (also known as sodium cromoglycate) and nedocromil (Tilade), are effective drugs in asthma management. These stabilizers hinder histamine release by skillfully obstructing the activation of mast cells and other cellular entities. Notably, they navigate this task without...

Drugs for Treatment of Crohn's Disease in IBD Using Glucocorticoids

293
Glucocorticoids, a class of anti-inflammatory drugs, are pivotal in treating moderate to severe Crohn's disease by inducing remission. They exhibit their anti-inflammatory action by inhibiting the production of inflammatory cytokines such as tumor necrosis factor (TNF)-α, interleukin (IL)-1, and chemokines like IL-8. In addition, they reduce the expression of inflammatory cell adhesion molecules and inhibit gene transcription of nitric oxide synthase, phospholipase A2, cyclooxygenase-2...

Area of Science:

  • Materials Science and Engineering
  • Pharmaceutical Sciences
  • Dermatology

Background:

  • Dissolving microneedles (DMN) offer minimally invasive transdermal drug delivery.
  • Low drug solubility limits therapeutic dosage in conventional DMN systems.
  • Triamcinolone acetonide (TA), a potent corticosteroid, is challenging to formulate into DMN due to poor solubility, hindering effective treatment for atopic dermatitis (AD).

Purpose of the Study:

  • To develop high-dose TA-loaded DMN (TA-DMN) overcoming solubility challenges.
  • To optimize fabrication methods for stable, high-concentration TA suspensions within DMN.
  • To evaluate the in vitro performance and in vivo anti-inflammatory efficacy of TA-DMN for AD treatment.

Main Methods:

  • Utilized sonication to reduce particle size and optimize polymer composition for dispersion stability of TA suspension.
  • Fabricated high-dose TA-DMN using optimized formulation and processing parameters.
  • Assessed in vitro physical characteristics and in vivo anti-inflammatory effects in a mouse model mimicking human AD.

Main Results:

  • Successfully fabricated high-dose TA-DMN by optimizing sonication and polymer composition.
  • Achieved good dispersion stability and particle size reduction of TA within the microneedles.
  • Demonstrated significant in vivo anti-inflammatory effects of TA-DMN in a skin inflammation mouse model.

Conclusions:

  • High-dose TA-DMN represent a promising strategy for effectively treating atopic dermatitis (AD).
  • This DMN formulation overcomes the limitations of poor drug solubility for potent corticosteroids.
  • TA-DMN have broad potential for treating inflammatory skin diseases requiring high-dose steroid therapy.
