Aneurysm II: Clinical Manifestations and Diagnostic Studies

84
Thoracic, aortic arch and abdominal aneurysms are significant vascular conditions that can present with various clinical manifestations and lead to serious complications. Understanding these manifestations and the appropriate diagnostic studies is essential for effective management and treatment.Thoracic Aortic AneurysmsThoracic aortic aneurysms often remain asymptomatic until they reach a size that impinges on adjacent structures. They typically cause deep, diffuse chest pain that radiates to...

Aneurysm I: Introduction

An aortic aneurysm is a localized outpouching or dilation at a weak point in the artery wall. It may involve different parts of the aorta, such as the abdominal aorta, aortic arch, or thoracic aorta.Etiological factorsSeveral disorders are associated with aortic aneurysms.Congenital causes, such as primary connective tissue disorders like Marfan syndrome, impact the integrity and strength of connective tissues, notably affecting the aorta. Marfan syndrome is a genetic disorder that specifically...

Area of Science:

  • Vascular Biology
  • Epigenetics
  • RNA Biology

Background:

  • Primary multiple intracranial aneurysm (MIA) is a dangerous vascular disease often leading to fatal rupture and subarachnoid hemorrhage.
  • The epigenetic mechanisms underlying MIA, particularly the role of circular RNAs (circRNAs), remain largely unexplored.
  • circRNAs are implicated in cardiovascular diseases, but their specific involvement in MIA requires investigation.

Purpose of the Study:

  • To explore novel molecular mechanisms in MIA by examining circRNA expression profiles.
  • To identify differentially expressed circRNAs and their potential regulatory roles in MIA pathogenesis.
  • To investigate the circRNA-microRNA-mRNA regulatory network in MIA.

Main Methods:

  • RNA sequencing (RNA-Seq) was performed on human peripheral blood mononuclear cells to detect comprehensive circRNA expression profiles.
  • Reverse transcription-quantitative PCR (RT-qPCR) was used to validate RNA-Seq findings.
  • Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analyses were conducted to determine the functions of identified circRNAs.

Main Results:

  • Out of 3,328 differentially expressed circRNAs, 60 showed significant changes in MIA patients compared to controls.
  • Forty downregulated circRNAs were identified, with several implicated in inflammatory processes.
  • The most significant KEGG pathway identified was 'leukocyte transendothelial migration', with specific circRNAs regulating key genes like PTK2, PRKCB, and ITGA.

Conclusions:

  • This study represents the first circRNA sequencing analysis in MIA, revealing a significant regulatory role for circRNAs in MIA pathogenesis.
  • Downregulated circRNAs, particularly those involved in inflammation and leukocyte transendothelial migration, are critical in MIA development.
  • The identified circRNAs and pathways provide novel insights into MIA etiology and potential therapeutic targets.
