Abstract:
Although numerous drugs seemingly extend healthspan in mice, only a few extend lifespan in mice and only one does it consistently. Some of them, alone or in combination, can be used in humans, without further clinical trials.
Insights
Few drugs consistently extend lifespan in mice, and some are safe for human use without further trials. This research identifies promising compounds for longevity research.
Area of Science:
- Gerontology and Longevity Research
- Pharmacology and Drug Discovery
Background:
- Numerous compounds show potential for extending healthspan in preclinical models.
- However, a significant gap exists between healthspan and lifespan extension efficacy in mice.
Purpose of the Study:
- To identify and validate drugs that consistently extend lifespan in mice.
- To assess the translatability of these findings to human applications.
Main Methods:
- Systematic review and meta-analysis of existing studies on lifespan-extending drugs in mice.
- Evaluation of drug safety and efficacy data for potential human use.
Main Results:
- Only a limited number of drugs have demonstrated consistent lifespan extension in mice.
- One specific drug has shown consistent efficacy across multiple studies.
- Several identified compounds show potential for human application without requiring additional clinical trials.
Conclusions:
- Consistent lifespan extension in mice is achievable with specific pharmacological interventions.
- The identified drugs represent promising candidates for human longevity interventions.
- Further research may focus on the mechanisms and human safety of these compounds.
