Summary

The Silk Fibroin/Polyurethanes (SF/PU) patch shows good compatibility in diabetic rats, with no serious complications. Fibrosis in diabetes promotes new vascular growth within the patch.

Background:

  • Diabetes mellitus (DM) poses challenges for biomaterial integration due to altered host responses.
  • Silk Fibroin/Polyurethanes (SF/PU) are investigated as potential vascular graft materials.

Purpose of the Study:

  • To evaluate the histopathological characteristics and host response to SF/PU patches in a diabetic rat model.
  • To assess the compatibility of SF/PU patches in a hyperglycemic environment.

Main Methods:

  • Diabetic (DM) and control (CT) rat groups were established.
  • SF/PU patches were implanted in the abdominal aorta and explanted at 1, 2, 3, and 4 weeks post-implantation.

Main Results:

  • Diabetic rats exhibited increased fibrosis and delayed endothelialization compared to controls.
  • No chronic inflammation was observed in either group.
  • Fibrosis in the DM group supported the formation of new vascular structures, including endothelial and smooth muscle cells.

Conclusions:

  • The SF/PU patch demonstrates suitability for implantation in diabetic individuals.
  • Hyperglycemia-induced fibrosis may promote beneficial vascularization within the SF/PU patch.
  • The SF/PU patch shows no significant complications in a hyperglycemic setting.
