Area of Science:

  • Plant genetics
  • Genomics
  • Molecular biology

Background:

  • Microsatellite (SSR) markers are crucial for gene mapping in rye, wheat-rye translocations, and triticale.
  • Existing rye chromosome maps have limited SSR loci, necessitating the integration of new markers.
  • Wheat EST-SSRs offer a potential source for novel SSR markers in rye.

Purpose of the Study:

  • To integrate wheat EST-SSRs (Expressed Sequence Tag-derived SSRs) into existing rye genetic maps.
  • To construct a comprehensive consensus rye microsatellite map.
  • To expand the available SSR markers for rye chromosome mapping.

Main Methods:

  • Utilized four rye mapping populations (P87/P105, N6/N2, N7/N2, N7/N6).
  • Employed CFE (EST-SSRs) primers for mapping.
  • Integrated mapping data to construct a consensus map.

Main Results:

  • Successfully mapped 23 Xcfe loci (wheat EST-SSRs) to specific rye chromosomes (1R-7R).
  • Identified orthologous positions for most mapped Xcfe loci relative to common wheat, considering evolutionary translocations.
  • Constructed a consensus rye microsatellite map comprising 123 SSRs, 12 SNPs, 118 RFLPs, and 2 isozyme loci.

Conclusions:

  • The integration of wheat EST-SSRs significantly enriches the rye genetic map.
  • The developed consensus map provides a valuable resource for gene mapping and QTL analysis in rye.
  • Orthologous SSR loci facilitate comparative genomics between wheat and rye.
