Purpose of the Study:

  • To identify epigenetic signatures linked to the diffusing capacity of the lung for carbon monoxide (D_LCO).

Main Methods:

  • Conducted an epigenome-wide association study (EWAS) in 2674 individuals from the Rotterdam Study and Framingham Heart Study.
  • Utilized the single-breath technique to measure D_LCO and D_LCO/V_A.
  • Assessed clinical relevance through whole blood and lung tissue gene expression analysis.

Main Results:

  • Identified and replicated two CpG sites (cg05575921, cg05951221) associated with D_LCO/V_A, and one (cg05575921) suggestively with D_LCO.
  • Found AHRR gene hypomethylation (cg05575921) positively correlated with EXOC3 gene expression in whole blood.
  • Confirmed EXOC3 expression in lung tissue positively correlates with D_LCO/V_A and D_LCO.

Conclusions:

  • Reported epigenome-wide associations with diffusing capacity in a general population.
  • EXOC3 is a strong candidate gene potentially mediating the effects of smoking-induced AHRR hypomethylation on pulmonary gas exchange.
