Summary
This summary is machine-generated.

Genomic tests Decipher and Prolaris show correlated scores for predicting prostatectomy outcomes. Decipher accurately predicted metastasis in a larger cohort, while Prolaris-derived scores did not.

Area of Science:

  • Urology
  • Genomics
  • Oncology

Background:

  • Multiple genomic tests exist post-radical prostatectomy (RP) but lack comparative evidence.
  • This study compares the predictive performance of two genomic tests for oncological outcomes after RP.

Purpose of the Study:

  • To compare the performance of Decipher (D) and Prolaris (P) genomic tests in predicting post-radical prostatectomy (RP) oncological outcomes.
  • To evaluate the correlation between D and P scores and their association with biochemical recurrence (BCR) and metastasis (M).

Main Methods:

  • Compared Decipher (D) scores with Prolaris (P) cell cycle progression (CCP) scores in 16 post-RP patients.
  • Derived a microarray CCP (mCCP) from D and correlated it with P-CCP.
  • Evaluated D and mCCP associations with BCR and M using multivariable survival analysis in a large RP cohort (1992-2010).

Main Results:

  • Significant correlation found between D scores and P-CCP (r=0.67, p=0.004).
  • Microarray CCP (mCCP) derived from D strongly correlated with P-CCP (r=0.88, p=6.7e-6).
  • In a larger cohort, Decipher (D) predicted metastasis (M) (HR 1.3, p=0.0005), while mCCP did not (p=0.62).

Conclusions:

  • Decipher and Prolaris scores are correlated and aid in identifying high-risk patients for BCR post-RP.
  • Decipher demonstrated predictive value for metastasis in a larger cohort, unlike the derived mCCP.
