The transcription factor CDX2 upregulates interleukin-33 (IL-33) gene expression in the intestines. This study provides the first evidence that CDX2 controls IL-33 levels, impacting inflammatory and autoimmune diseases.
Area of Science:
- Molecular Biology
- Genetics
- Immunology
Background:
- Interleukin-33 (IL-33) dysregulation is linked to autoimmune and inflammatory diseases.
- The transcriptional regulation of the IL33 gene remains largely unexamined.
- CDX2 is a key intestinal transcription factor regulating gene networks in the gut.
Purpose of the Study:
- To investigate the role of the intestinal transcription factor CDX2 in regulating IL33 gene expression.
- To determine if CDX2 influences IL33 mRNA levels in colonic epithelial cells.
Main Methods:
- Analysis of IL33 mRNA expression in primary human colonic epithelial cells and cell lines.
- Genomic analyses including ChIP-seq and RNA-seq in LS174T cells.
- Promoter analysis, Western blotting, and qRT-PCR to assess CDX2-dependent IL33 regulation.
Main Results:
- High IL33 mRNA expression was observed in primary colonic epithelial cells and LS174T cells.
- Genomic data revealed CDX2-dependent intronic enhancer activity within the IL33 gene.
- IL33 expression was confirmed to be upregulated in a CDX2 concentration-dependent manner.
Conclusions:
- CDX2 directly regulates IL33 gene expression.
- This finding provides the first evidence of CDX2 controlling IL33 transcription.
- Understanding this regulatory mechanism may offer insights into IL-33-associated inflammatory diseases.
