Summary

Regulatory T (Treg) cells in human tissues were defined by a novel follicular helper T cell-like signature. This signature, governed by BATF, is linked to tissue repair functions and CCR8 expression.

Area of Science:

  • Immunology
  • Cell Biology
  • Human Tissues

Background:

  • Regulatory T (Treg) cells play crucial roles in maintaining immune homeostasis.
  • Defining tissue-specific Treg cell subsets is essential for understanding their functions in health and disease.

Purpose of the Study:

  • To characterize the molecular signature of Treg cells in healthy human tissues.
  • To identify novel Treg cell subsets and their functional attributes.

Main Methods:

  • Analysis of chromatin accessibility data.
  • Transcriptome and protein-level analysis.
  • Identification of key transcription factors and cell surface markers.

Main Results:

  • A distinct human follicular helper T cell-like Treg cell signature was identified.
  • The transcription factor BATF governs this Treg cell subset.
  • This signature is associated with tissue repair functions and CCR8 expression.

Conclusions:

  • A novel tissue-repair Treg cell subset is defined by a follicular helper T cell-like signature.
  • BATF is a key regulator of this Treg cell population.
  • CCR8 expression serves as a functional marker for these tissue-repair Treg cells.
