Myelin basic protein (Mbp) promotes axonal regeneration from neural progenitor cells (NPCs) via a novel Mbp/L1cam/Pparγ pathway. This discovery offers potential for bioengineered NPC therapies to enhance recovery after spinal cord injury (SCI).
Area of Science:
- Neuroscience
- Regenerative Medicine
- Biochemistry
Background:
- Neural progenitor cell (NPC) therapy shows promise for spinal cord injury (SCI) recovery.
- Myelin presence stimulates axonal regeneration from mammalian NPCs.
- Myelin-associated proteins are hypothesized to contribute to NPC-mediated axonal regeneration.
Purpose of the Study:
- To identify key genes involved in myelin-associated axonal regeneration from NPCs.
- To investigate the role of myelin basic protein (Mbp) in NPC axonal regeneration.
- To elucidate the signaling pathways mediating Mbp's effects on NPC neurite outgrowth and in vivo SCI recovery.
Main Methods:
- Bioinformatic analysis identified Mbp as a key gene.
- Utilized murine, rat, and human iPSC-derived NPCs with Mbp knockout/overexpression and pathway modulators.
- Assessed in vivo locomotive recovery and axonal regeneration in a murine dorsal column crush SCI model using engineered NPCs on scaffolding.
Main Results:
- Mbp promotes NPC axonal outgrowth in a Mbp-dependent manner, mediated by L1-70.
- Mbp/L1-70 stimulates NPC neurite outgrowth via PPARγ-mediated repression of differentiation genes and Erk1/2 activation.
- In vivo, Mbp-overexpressing NPCs significantly improved locomotive recovery and axonal regeneration in SCI mice.
Conclusions:
- Mbp supports mammalian NPC axonal regeneration through the novel Mbp/L1cam/Pparγ signaling pathway.
- Bioengineered NPC interventions hold potential for promoting axonal regeneration and functional recovery post-SCI.
