Summary

Heavy cannabis use (HCU) is linked to sensorimotor deficits and altered brain structure. This study found higher neurological soft signs (NSS) and gray matter volume (GMV) differences in the brains of HCU individuals compared to controls.

Background:

  • Sensorimotor dysfunction and neurological soft signs (NSS) are documented in cannabis dependence.
  • It remains unclear if heavy cannabis use (HCU) without dependence also leads to sensorimotor abnormalities and their neural underpinnings.

Purpose of the Study:

  • To investigate the association between NSS and gray matter volume (GMV) in males with HCU compared to controls.
  • To identify specific sensorimotor deficits and their corresponding neural correlates in HCU.

Main Methods:

  • Structural magnetic resonance imaging (MRI) and voxel-based morphometry (VBM) were used to assess GMV in 24 HCU males and 17 controls.
  • Standardized NSS assessments were performed to evaluate sensorimotor performance across different domains.

Main Results:

  • Individuals with HCU exhibited significantly higher total NSS scores, particularly in motor coordination, complex motor tasks, and hard signs.
  • HCU was associated with significant NSS/GMV interactions in regions including the putamen, inferior frontal cortex, cerebellum, precentral cortex, and thalamus.
  • Between-group analyses revealed lower GMV in the right anterior orbital and precentral gyrus, and higher GMV in the right superior frontal gyrus and left supplementary motor cortex in HCU individuals.

Conclusions:

  • The findings support the notion that heavy cannabis use is associated with abnormal sensorimotor performance.
  • Aberrant cortical-thalamic-cerebellar-cortical circuits may underlie the observed sensorimotor deficits in individuals with HCU.
