Summary

Nitroso-2-hydroxyethylurea (NHEU) causes spleen hemangiosarcomas in hamsters. Removing the spleen prevented liver tumors, confirming they were metastases, not primary growths.

Purpose of the Study:

  • To determine if liver hemangiosarcomas induced by nitroso-2-hydroxyethylurea (NHEU) are true metastases from the spleen.
  • To evaluate the effect of splenectomy on NHEU-induced carcinogenesis.
  • To assess the specificity of NHEU's carcinogenic activity.

Main Methods:

  • Surgically splenectomized female Syrian hamsters were treated with NHEU weekly for 22 weeks.
  • A control group of unsplenectomized hamsters received identical NHEU treatment.

    • Splenectomized hamsters survived significantly longer (median 45 weeks) than unsplenectomized hamsters (median 24 weeks).
