Sleep is an essential physiological process vital to maintaining overall well-being. The reticular activating system (RAS), a network of neurons in the brainstem, regulates wakefulness and sleep. While it may seem passive, sleep consists of distinct cycles, each with its unique characteristics and functions. Two key sleep phases are non-rapid eye movement (NREM) and  rapid eye movement (REM).

Background:

  • Pannexin proteins are homologous to invertebrate gap junction proteins.
  • Six variants of the PANX1 gene are linked to human infertility and oocyte defects.
  • Previous Panx1 knockout mouse models showed mild phenotypes and contained confounding mutations.

Purpose of the Study:

  • To create new, precise mouse models for studying PANX1.
  • To re-evaluate the function of PANX1 and the Arg217His variant in vivo.
  • To investigate previously reported effects of Panx1 knockout on mouse behavior.

Main Methods:

  • CRISPR/Cas9 gene editing was used to generate Panx1 knockout mice.
  • CRISPR/Cas9 was used to create mice with the human Arg217His PANX1 substitution.
