Area of Science:

  • Neuroscience
  • Developmental Biology
  • Comparative Anatomy

Background:

  • Mammalian neocortex size and shape vary significantly across species.
  • Understanding neocortical development is crucial for comparative studies.

Purpose of the Study:

  • To describe three ex vivo neocortical slice/tissue culture methods.
  • To provide techniques for analyzing neocortical development across species.

Main Methods:

  • Organotypic slice culture (mouse, ferret, human).
  • Hemisphere rotation culture (mouse, ferret).
  • Free-floating tissue culture (mouse, ferret, human).

Main Results:

  • Each method offers distinct analytical capabilities.
  • Cultures can be combined with genetic manipulation (electroporation) and inhibitor treatments.

Conclusions:

  • These culture methods are powerful tools for studying neocortical development.
  • They facilitate the examination of gene functions in mammalian brain development.
