Cholinergic Receptors: Muscarinic01:25

Cholinergic Receptors: Muscarinic

3.9K
The pharmacological actions of acetylcholine are elicited via its binding to two families of cholinergic receptors or cholinoceptors, namely, muscarinic and nicotinic receptors. Muscarinic receptors are G protein-coupled receptors and have five subtypes, M1–M5. All mAChR subtypes are activated by acetylcholine and blocked by the antagonist, atropine.
The subtypes M1, M3, and M5 couple with the Gq subunit and activate the phospholipase C (PLC) activity, mobilizing intracellular Ca2+....

Summary

Cholestatic itch, a severe symptom of liver disease, may be caused by bile acids and bilirubin acting on the Mas-related G protein-coupled receptor X4 (MRGPRX4). This discovery offers a new target for developing effective anti-itch therapies.

Area of Science:

  • Hepatology
  • Dermatology
  • Pharmacology

Background:

  • Pruritus (itch) is a debilitating symptom in cholestatic liver diseases, significantly impairing patient quality of life.
  • Current treatment options for cholestatic itch are limited due to an incomplete understanding of its molecular underpinnings.
  • Potential pruritogens include bile acids, bilirubin, lysophosphatidic acid, and endogenous opioids.

Purpose of the Study:

  • To review current knowledge on cholestatic itch mechanisms.
  • To highlight the role of Mas-related G protein-coupled receptor X4 (MRGPRX4) as a receptor for bile acids and bilirubin in mediating cholestatic itch.
  • To discuss future research directions and therapeutic targets for cholestatic itch.

Main Methods:

  • Literature review summarizing existing research on cholestatic itch.
  • Focus on recent findings identifying MRGPRX4 as a key receptor.
  • Discussion of proposed molecular mechanisms and therapeutic implications.

Main Results:

  • Mas-related G protein-coupled receptor X4 (MRGPRX4) has been identified as a receptor for bile acids and bilirubin.
  • MRGPRX4 is implicated as a mediator of cholestatic itch in humans.
  • This identification provides a novel molecular target for anti-itch interventions.

Conclusions:

  • MRGPRX4 represents a significant breakthrough in understanding cholestatic itch.
  • Targeting MRGPRX4 offers a promising strategy for developing novel treatments for pruritus in liver disease.
  • Further research is warranted to fully elucidate the role of MRGPRX4 and develop targeted therapies.
