Summary

Irradiating platelet concentrates does not harm their function or increase plasticizer leaching, even after 5 days of storage. This finding supports extended storage for platelet products, benefiting blood banks and patient care.

Purpose of the Study:

  • To assess the impact of gamma irradiation on in vitro platelet function.
  • To evaluate the leaching of plasticizers from storage bags post-irradiation.
  • To determine the feasibility of extended storage for irradiated platelet concentrates.

Main Methods:

  • Platelet concentrates (random-donor and apheresis) were irradiated with 2000 rads.
  • In vitro platelet function tests (count, aggregation, serotonin uptake) were performed immediately and after 5 days.
  • Plasticizer (DEHP) and its metabolite (MEHP) levels in bags were quantified.

  • Irradiation did not alter platelet count, white cell count, pH, glucose, or lactate levels.
  • Platelet aggregation, release reaction, and serotonin uptake remained unaffected by irradiation.
  • Irradiation did not increase the leaching of di(2-ethylhexyl)phthalate (DEHP) or its conversion to mono(2-ethylhexyl)phthalate (MEHP).

Conclusions:

  • Platelet concentrates can be irradiated on the day of collection.
  • Irradiated platelet concentrates maintain in vitro function for at least 5 days.
  • This extended storage capability offers significant advantages for blood bank logistics and product availability.
