Sestrins protect muscle stem cells (MuSCs) from aging by maintaining their quiescent state. Loss of Sestrins leads to premature stem cell activation and impaired muscle regeneration in aged mice.
Area of Science:
- Cellular biology
- Stem cell research
- Aging research
Background:
- Skeletal muscle health relies on quiescent muscle stem cells (MuSCs).
- Aging and metabolic stress impair MuSC quiescence, leading to exhaustion.
- Sestrins are stress-inducible proteins with antioxidant and mTORC1-inhibiting functions.
Purpose of the Study:
- To investigate the role of Sestrins in maintaining adult muscle stem cell quiescence.
- To determine the impact of Sestrins on mTORC1 signaling and metabolism in MuSCs.
Main Methods:
- Utilized SESTRIN1,2 knockout mice.
- Analyzed mTORC1 complex activity, cell cycle propensity, and metabolic flux.
- Assessed MuSC populations and muscle regeneration capacity in aged knockout mice.
Main Results:
- SESTRIN1,2 loss caused mTORC1 hyperactivation and increased cell cycle entry in MuSCs.
- Mice lacking Sestrins showed altered metabolic flux within MuSCs.
- Aged SESTRIN1,2 knockout mice exhibited significant MuSC loss and impaired muscle regeneration.
Conclusions:
- Sestrins are crucial for preserving muscle stem cell quiescence.
- Sestrins maintain essential metabolic pathways that protect MuSCs against aging-related decline.
- Targeting Sestrins may offer therapeutic strategies for age-related muscle degeneration.
