Non-LTR Retrotransposons03:18

Non-LTR Retrotransposons

12.2K
As the name suggests, non-LTR retrotransposons lack the long terminal repeats characteristic of the LTR retrotransposons. Additionally, both LTR and non-LTR retrotransposons use distinct mechanisms of mobilization. Non-LTR retrotransposons are further divided into two classes - Long interspersed nuclear elements (LINEs) and short interspersed nuclear elements (SINEs), both of which occur abundantly in most mammals, including humans. Some of the active non-LTR retrotransposons in humans are L1...

Abnormal Proliferation02:23

Abnormal Proliferation

4.7K
Under normal conditions, most adult cells remain in a non-proliferative state unless stimulated by internal or external factors to replace lost cells. Abnormal cell proliferation is a condition in which the cell's growth exceeds and is uncoordinated with normal cells. In such situations, cell division persists in the same excessive manner even after cessation of the stimuli, leading to persistent tumors. The tumor arises from the damaged cells that replicate to pass the damage to the...

Summary

DNA-methyltransferase 1 (DNMT1) mRNA is elevated in leukemia exosomes and can transfer to target cells. This transfer may reprogram cells, influencing leukemia progression and epigenetic changes.

Area of Science:

  • Biochemistry
  • Molecular Biology
  • Cancer Research

Background:

  • DNA-methyltransferase 1 (DNMT1) is crucial for embryonic development.
  • Aberrant DNMT1 expression drives tumor and leukemia progression via epigenetic alterations.
  • Exosomes play a role in intercellular communication and disease progression.

Purpose of the Study:

  • To investigate DNMT1 mRNA levels in exosomes derived from leukemia.
  • To determine if exosomal DNMT1 mRNA can be transferred to target cells.
  • To explore the potential role of exosomal DNMT1 in leukemia reprogramming.

Main Methods:

  • Quantitative analysis of DNMT1 mRNA in exosomes (Exo-PALL, Exo-HD, Exo-CM).
  • Co-culture experiments using leukemia B cells as target cells.
  • Detection of exosomal DNMT1 mRNA transfer into target cells.

Main Results:

  • DNMT1 mRNA transcripts were significantly elevated in Exo-PALL compared to Exo-HD.
  • Heightened DNMT1 mRNA levels were confirmed in exosomes from leukemia cell lines (Exo-CM).
  • Transfer of exosomal DNMT1 mRNA into target leukemia B cells was observed.

Conclusions:

  • Exosomes from leukemia cells carry elevated levels of DNMT1 mRNA.
  • Exosomal DNMT1 mRNA can be transferred to recipient leukemia cells.
  • This transfer mechanism may contribute to leukemia progression by reprogramming cellular epigenetics.
