Regulation of Angiogenesis and Blood Supply01:24

Regulation of Angiogenesis and Blood Supply

2.9K
Rapidly dividing tumors, embryos, and wounded tissues require more oxygen than usual, lowering the oxygen concentration in the blood. At low oxygen or hypoxic conditions, an oxygen-sensitive transcription factor called the hypoxia-inducible factor 1 or HIF1 is activated. HIF1 is a dimeric protein of alpha (ɑ) and beta (β) subunits.  Under optimal oxygen conditions, HIF1β is present in the nucleus while HIF1ɑ remains in the cytosol. HIF1ɑ is hydroxylated by prolyl...

Pleiotropy01:33

Pleiotropy

41.6K
Pleiotropy is the phenomenon in which a single gene impacts multiple, seemingly unrelated phenotypic traits. For example, defects in the SOX10 gene cause Waardenburg Syndrome Type 4, or WS4, which can cause defects in pigmentation, hearing impairments, and an absence of intestinal contractions necessary for elimination. This diversity of phenotypes results from the expression pattern of SOX10 in early embryonic and fetal development. SOX10 is found in neural crest cells that form melanocytes,...

Area of Science:

  • Neuroscience
  • Developmental Biology
  • Molecular Biology

Background:

  • The choroid plexus is a vital blood barrier producing cerebrospinal fluid, essential for brain development and homeostasis.
  • The OTX2 homeoprotein is critical for choroid plexus development and remains highly expressed in adults.

Purpose of the Study:

  • To investigate the functional roles of OTX2 in adult choroid plexus function.
  • To identify OTX2-regulated genes and protein interactions within the choroid plexus and its target regions.

Main Methods:

  • RNA sequencing analysis of OTX2 knockdown mouse models.
  • Mass spectrometry to identify OTX2 protein interactors.
  • Analysis of OTX2's impact on gene expression, splicing, and protein targets.

Main Results:

  • OTX2 regulates cell signaling, adhesion, and the expression of secreted factors like transthyretin in the choroid plexus.
  • OTX2 influences choroid plexus immune and stress responses, including oxidative stress and DNA repair pathways.
  • Identified OTX2 protein partners involved in cell adhesion, chromatin structure, and RNA processing.

Conclusions:

  • OTX2 plays significant roles in maintaining adult choroid plexus function and overall brain homeostasis.
  • OTX2's regulatory functions extend to immune responses, stress, and RNA processing, highlighting its lifelong importance.
