Targeting CD146 (melanoma cell adhesion molecule) shows promise for treating neuroblastoma, a common childhood cancer. Inhibiting CD146 effectively reduced tumor growth and increased cancer cell death in preclinical studies.
Area of Science:
- Oncology
- Molecular Biology
- Cancer Therapeutics
Background:
- Neuroblastoma is the most common pediatric extracranial solid tumor.
- High-risk or recurrent neuroblastoma has a poor prognosis despite current therapies.
- Novel therapeutic strategies are urgently needed for neuroblastoma treatment.
Purpose of the Study:
- To investigate the expression of CD146 (melanoma cell adhesion molecule) in neuroblastoma.
- To evaluate the anti-tumor effects of targeting CD146 in neuroblastoma cells.
- To explore CD146 as a potential therapeutic target for neuroblastoma.
Main Methods:
- Assessed CD146 expression in neuroblastoma cell lines and clinical samples.
- Utilized short hairpin RNA (shRNA) to knock down CD146 expression.
- Administered an anti-CD146 polyclonal antibody for in vitro and in vivo studies.
- Investigated apoptosis and signaling pathways (FAK, NF-κB).
Main Results:
- CD146 was expressed in neuroblastoma cell lines and primary tumors.
- CD146 knockdown or antibody treatment inhibited neuroblastoma cell growth in vitro.
- Anti-CD146 antibody treatment significantly reduced tumor growth in vivo.
- Inhibition of CD146 led to increased apoptosis via FAK and/or NF-κB pathways.
Conclusions:
- CD146 is expressed in neuroblastoma and serves as a potential therapeutic target.
- Targeting CD146 demonstrates significant anti-tumor activity against neuroblastoma.
- CD146-targeting therapies warrant further investigation for neuroblastoma treatment.
