Psychologists have long debated whether language shapes thought or vice versa. This discussion gained grip with Edward Sapir and Benjamin Lee Whorf in the 1940s, who proposed that language determines thought, a concept known as linguistic determinism. They suggested that the vocabulary and structure of a language influence how its speakers think and perceive reality.

  • Diffusion MRI data from 236 participants (cognitively unimpaired and mild cognitive impairment) were analyzed using the free-water elimination technique.

  • General linear modeling and competitive models assessed relationships between microstructural values, SCD scores, and cerebrospinal fluid (CSF) amyloid-β (Aβ42).

  • CSF Aβ42 was associated with SCD, but specific white matter tracts (fornix, IFG pars triangularis) uniquely predicted SCD beyond amyloid levels.
  • Tracts like the inferior longitudinal fasciculus (ILF) RD_T, fornix RD_T, and uncinate fasciculus (UF) FW showed the strongest association with SCD scores.

Conclusions:

  • Medial temporal lobe and frontal transcallosal tract microstructure significantly drives SCD scores, independent of early amyloid deposition.
  • Abnormal white matter diffusivity represents a crucial early contributor to cognitive decline.
  • Integrating multiple biomarkers, including white matter microstructure, is vital for understanding SCD heterogeneity.
