Colon cancer growth and glycolysis are promoted by karyopherin α2 subunit (KPNA2). Centromere protein A (CENPA) activates KPNA2 by recruiting GCN-5, driving cancer progression.
Area of Science:
- Oncology
- Molecular Biology
- Cancer Metabolism
Background:
- The karyopherin α2 subunit gene (KPNA2) is an oncogene implicated in cancer metabolic reprogramming.
- Understanding KPNA2's role in colon cancer (CC) growth and glycolysis is crucial for targeted therapies.
Purpose of the Study:
- To investigate the function of KPNA2 in colon cancer cell growth and glycolysis.
- To identify upstream regulators of KPNA2 in colon cancer.
Main Methods:
- Bioinformatic screening of the Oncomine database for differentially expressed genes in CC.
- Experimental validation of KPNA2 expression in CC cell lines.
- Assessment of KPNA2 knockdown effects on cell viability, DNA replication, apoptosis, glucose consumption, extracellular acidification rate, and ATP production.
- Identification and validation of Centromere protein A (CENPA) as an upstream activator of KPNA2, involving GCN-5 and H3K27ac modification.
Main Results:
- KPNA2 was found to be highly expressed in colon cancer and CC cell lines.
- Down-regulation of KPNA2 significantly reduced colon cancer cell viability, DNA replication, glucose consumption, extracellular acidification rate, and ATP production, while increasing apoptosis.
- Centromere protein A (CENPA) was identified as an upstream transcription activator of KPNA2, recruiting GCN-5 to the KPNA2 promoter, leading to transcriptional activation.
- Overexpression of CENPA or GCN-5 counteracted the effects of KPNA2 knockdown, restoring growth and glycolysis.
Conclusions:
- The study elucidates a novel regulatory pathway where CENPA recruits GCN-5 to activate KPNA2 transcription.
- Activated KPNA2 enhances colon cancer cell growth and glycolysis, contributing to tumor development.
- This pathway represents a potential therapeutic target for colon cancer treatment.
