Area of Science:

  • Virology
  • Molecular Diagnostics
  • Infectious Disease Research

Background:

  • The rapid global spread of SARS-CoV-2 necessitated the development of accurate and swift diagnostic tests.
  • Numerous commercial platforms for rapid virus detection have emerged and received approval.

Purpose of the Study:

  • To evaluate the performance of four commercial RT-qPCR kits and two other diagnostic systems for SARS-CoV-2 detection.
  • To compare the sensitivity and reliability of different commercial assays using clinical samples and serial dilutions.

Main Methods:

  • Analysis of 95 positive and 5 negative retrospective clinical samples.
  • Testing of four RT-qPCR kits: TaqMan 2019nCoV Assay, Allplex™SARS-COV-2 Assay, FTD SARS-COV-2 Assay, and qCOVID-19.
  • Evaluation of Hologic Aptima SARS-COV-2 and Clart-COVID-19 systems, including sensitivity analysis with SARS-CoV-2 standard dilutions.

Main Results:

  • qCOVID-19 and Allplex™SARS-COV-2 Assay detected all clinical samples.
  • All four RT-qPCR kits demonstrated high sensitivity for samples with Ct<33.
  • Clart-COVID-19 microarrays successfully detected all samples and controls, while Hologic Aptima Panther failed with one sample and had numerous invalidated results.

Conclusions:

  • All evaluated diagnostic techniques are valuable for SARS-CoV-2 detection.
  • Specific RT-qPCR assays like qCOVID-19 and Allplex™SARS-COV-2 Assay showed excellent performance in detecting clinical samples.
  • The Clart-COVID-19 system demonstrated reliable detection, whereas the Hologic Aptima Panther system presented challenges with sample validation.
