Area of Science:

  • Virology
  • Epidemiology
  • Regulatory Science

Background:

  • Filoviruses, including Sudan ebolavirus and Marburg virus, pose significant public health threats, necessitating the development of medical countermeasures.
  • While Zaire ebolavirus has licensed vaccines and therapeutics, other filoviruses lack approved treatments.
  • Development of countermeasures for less common filoviruses often relies on nonclinical studies under the U.S. Food and Drug Administration (FDA) Animal Rule.

Purpose of the Study:

  • To recommend appropriate virus challenge isolates for Sudan ebolavirus and Marburg virus nonclinical studies.
  • To guide the selection of prototype virus strains for regulatory submissions under the FDA Animal Rule.
  • To facilitate the development of effective medical countermeasures for Sudan ebolavirus and Marburg virus.

Main Methods:

  • Analysis of existing data and considerations for strain selection under the FDA Animal Rule Guidance.
  • Led by the Filovirus Animal and Nonclinical Group (FANG), this study focused on identifying suitable challenge isolates.
  • Evaluation of virus isolates for their relevance in nonclinical challenge studies.

Main Results:

  • Proposed prototype virus isolates for Sudan ebolavirus and Marburg virus.
  • Provided recommendations for selecting challenge isolates crucial for nonclinical studies.
  • Established a basis for future regulatory submissions for Sudan ebolavirus and Marburg virus countermeasures.

Conclusions:

  • The selection of appropriate virus challenge isolates is critical for the successful development of medical countermeasures against Sudan ebolavirus and Marburg virus.
  • The proposed prototype isolates will aid in standardizing nonclinical studies and supporting regulatory approval.
  • This work supports the advancement of public health preparedness against filovirus threats.
