    Background:

    • Human corneal stromal fibroblast cultures were established from normal corneas.
    • Antigen expression profiles were analyzed before and after specific immune factor treatments.

    Purpose of the Study:

    • To investigate the expression of various antigens on human corneal fibroblasts.
    • To determine the effect of immune modulators like interferons and interleukins on antigen expression.
    • To understand the role of inducible antigen expression in corneal allograft rejection.

    Main Methods:

    • Primary human corneal fibroblast cultures were used.
    • Cells were treated with gamma interferon, alpha interferon, interleukin 2, and PMA.
    • Antigen expression (Class I, Class II, monocyte/granulocyte, T-cell) was assessed using immunophenotyping.

    Main Results:

    • Class I and monocyte/granulocyte antigens were constitutively expressed.
    • Class II (HLA-DR) antigens were induced by gamma interferon.
    • PMA reduced Leu M-1 positive cells; alpha IFN and IL-2 had no effect on antigen expression.
    • HLA-DR induction was rapid (12 hours) and sustained after gamma interferon removal.

    Conclusions:

    • Gamma interferon induces Class II antigen expression on corneal fibroblasts.
    • This inducible expression of Class II antigens may be critical in the immune response to corneal allografts.
    • Findings suggest a mechanism for immune rejection in corneal transplantation.
