Exercise and irisin injections benefit brain health by altering protein expression. Irisin may substitute for exercise when physical activity is not possible, offering a novel therapeutic approach for neurodegenerative diseases.
Area of Science:
- Neuroscience
- Exercise Physiology
- Molecular Biology
Background:
- Physical inactivity is linked to neurodegenerative diseases.
- Exercise offers significant neuroprotective benefits, particularly for cognitive function in older adults.
- Irisin is identified as a key mediator of exercise's positive effects on the brain.
Purpose of the Study:
- To investigate proteome alterations in mouse brain tissue following endurance exercise, resistance exercise, and irisin injection.
- To quantify irisin levels in blood.
- To compare the molecular effects of exercise modalities and irisin administration.
Main Methods:
- National Maritime Research Institute (NMRI) mice were subjected to endurance exercise, resistance exercise, or irisin injection.
- Blood irisin levels were quantified using an irisin-ELISA Kit.
- Brain tissue proteome was analyzed using two-dimensional gel electrophoresis and MALDI-TOF MS/MS.
Main Results:
- At least 21 proteins showed altered expression across the different treatment groups.
- Cellular pathway analysis indicated shared beneficial effects between irisin treatment and exercise.
- Irisin levels in blood were quantified.
Conclusions:
- Irisin injection demonstrates comparable beneficial effects to exercise on brain proteome.
- Irisin may serve as a potential therapeutic substitute for exercise, especially for individuals unable to engage in physical activity.
- This suggests a novel strategy for mitigating neurodegenerative disease risk.
