Purpose of the Study:

  • To investigate topological changes in the functional brain networks of panic disorder patients using a graph-theoretical approach.
  • To identify network-based biomarkers for panic disorder.

Main Methods:

  • Resting-state functional magnetic resonance imaging (fMRI) was performed on 31 patients with panic disorder and 33 healthy controls.
  • Functional brain networks were constructed using the Anatomical Automatic Labeling template and connectivity matrices.
  • Network-based statistical analysis was used to compare global and nodal network properties between groups.

Main Results:

  • Panic disorder patients exhibited a small-world network topology, differing from controls.
  • Patients showed reduced nodal efficiency in the right superior frontal gyrus (SFG), middle frontal gyrus (MFG), right superior temporal gyrus (STG), and left middle temporal gyrus (MTG).
  • Decreased functional connectivity was observed between the right middle temporal gyrus (MTG) and other temporal regions. Reduced SFG nodal efficiency correlated positively with symptom severity, while reduced angular gyrus betweenness centrality correlated negatively.

Conclusions:

  • Panic disorder is characterized by impaired global and regional information transmission within the brain's functional networks.
  • Temporal lobe regions play a significant role in the pathophysiology of panic disorder.
  • Network analysis reveals potential neuroimaging biomarkers for panic disorder.
