Spermatogenesis is the process by which haploid sperm cells are produced in the male testes. It starts with stem cells located close to the outer rim of seminiferous tubules. Background:

  • Spermatogonial stem cell transplantation is a method for producing donor-derived sperm in various animal species.
  • Germ cell transplantation has not been previously applied to stallions.
  • This study aimed to establish donor germ cell-derived sperm production in stallions via transplantation.

Observation:

  • Donor germ cells were transplanted into the testicular parenchyma of three recipient stallions treated with busulfan.
  • Semen was collected 8 weeks post-transplantation for sperm evaluation and genotyping.
  • Sperm motility remained above 50%, and stallion libido was unaffected.

Findings:

  • Microsatellite fingerprinting failed to detect any donor-derived sperm in the recipient stallions' semen.
  • The transplantation technique into the testicular parenchyma was ineffective for producing donor-derived sperm in stallions.

Implications:

  • Direct transplantation into the stallion testicular parenchyma is not a viable method for producing donor-derived sperm.
  • Further research is needed to explore alternative germ cell transplantation techniques in stallions.
  • This study highlights challenges in applying established stem cell technologies to new species.
