Summary

Brain perivascular macrophages do not mediate sickness behavior induced by interleukin-1beta (IL-1β). Targeting these cells with IL-1β-saporin did not alter IL-1β-induced behavioral changes in rats.

Background:

  • Sickness behavior, a reduction in activity, often follows bacterial infection or administration of lipopolysaccharide (LPS) and pro-inflammatory cytokines like interleukin-1beta (IL-1β).
  • Brain perivascular macrophages, along with microglia, are immune cells within the brain that express the IL-1 type 1 receptor (IL-1R1) and may contribute to sickness behavior.

Purpose of the Study:

  • To investigate the role of brain perivascular macrophages in mediating IL-1β-induced sickness behavior in rats.
  • To determine if eliminating IL-1R1-expressing brain cells affects sickness responses to IL-1β.

Main Methods:

  • Rats received intracerebroventricular (icv) administration of an IL-1β-saporin conjugate to eliminate IL-1R1-expressing cells, specifically targeting brain perivascular macrophages.
  • Following treatment, rats were administered IL-1β either centrally (icv) or peripherally (intraperitoneally, ip) to assess sickness behaviors.

Main Results:

  • Icv IL-1β-saporin administration successfully reduced the number of brain perivascular macrophages.
  • This reduction did not alter the subsequent sickness behaviors induced by either central or peripheral IL-1β administration, including decreased food intake, locomotor activity, and social interaction.

Conclusions:

  • Intracerebroventricular IL-1β-saporin administration is an effective method for targeting and reducing brain perivascular macrophages.
  • The study concludes that brain perivascular macrophages are not critically involved in mediating IL-1β-induced sickness behavior in rats.
