Enzyme-Linked Immunosorbent Assay

15.0K
In 1971, Peter Perlman and Eva Engvall developed an Enzyme-linked immunosorbent assay (ELISA or EIA). ELISA differs from western blot in that the assays are conducted in microtiter plates or in vivo rather than on an absorbent membrane.
There are many different types of ELISAs, but they all involve an antibody molecule whose constant region binds an enzyme, leaving the variable region free to bind its specific antigen.  Enzyme-substrate reaction allows the antigen to be visualized or...

Background:

  • Serology tests are crucial for identifying past pathogen exposure.
  • Measuring anti-SARS-CoV-2 antibodies is vital for understanding COVID-19 transmission dynamics.
  • High-throughput serological assays are needed for pandemic response.

Purpose of the Study:

  • To adapt Luminex bead-based technology for a COVID-19 multi-antigen serological assay.
  • To establish and optimize a protocol for measuring SARS-CoV-2 antibodies in human samples.
  • To provide a flexible assay for broad applicability in pathogen serosurveillance.

Main Methods:

  • Utilized Luminex bead-based technology for multiplexed antigen detection.
  • Developed and optimized a protocol for anti-SARS-CoV-2 antibody measurement.
  • Validated the assay for use with both plasma and saliva samples.

Main Results:

  • Successfully adapted Luminex technology for COVID-19 antibody detection.
  • Established a detailed protocol for assay optimization and implementation.
  • Demonstrated the assay's utility in measuring antibodies from different sample types.

Conclusions:

  • The developed Luminex-based assay is effective for high-throughput COVID-19 serology.
  • The protocol provides a robust method for SARS-CoV-2 antibody quantification.
  • This adaptable assay can be applied to study antibodies against various pathogens.
