Purpose of the Study:

  • To evaluate the consistency of metabarcoding results between the DNBSEQ-G400RS and NovaSeq 6000 platforms.
  • To compare the performance of DNA nanoball sequencing versus sequencing-by-synthesis for ecological studies.

Main Methods:

  • Technical replicates of COI gene amplicons from 120 soil DNA samples were sequenced on both DNBSEQ-G400RS and NovaSeq 6000.
  • Raw sequencing data underwent uniform bioinformatics processing to analyze operational taxonomic unit (OTU) and amplicon sequence variant (ASV) richness and composition.

Main Results:

  • Both platforms yielded a similar proportion of high-quality reads.
  • Per-sample OTU and ASV richness patterns were highly correlated, though DNBSEQ-G400RS showed higher OTU counts.
  • No statistical difference in ASV richness was observed after denoising, indicating effective noise removal.
  • OTU- and ASV-based community compositions were strongly correlated and largely interchangeable between platforms.

Conclusions:

  • DNBSEQ-G400RS and NovaSeq 6000 are equally efficient for high-throughput metabarcoding.
  • The DNBSEQ-G400RS platform offers a significant advantage in terms of lower sequencing cost.
