This meta-analysis found that the PPAR-γ rs1801282 C/G gene polymorphism is associated with an increased risk of ischemic stroke (IS). Further research is needed to confirm these findings on PPAR-γ and IS development.

Background:

  • Emerging research suggests a link between Peroxisome proliferator-activated receptor gamma (PPAR-γ) and the development of ischemic stroke (IS).

Purpose of the Study:

  • To conduct a meta-analysis evaluating the association between specific PPAR-γ gene polymorphisms (rs1801282 C/G and rs3856806 C/T) and the risk of developing IS.
  • To synthesize existing evidence to provide a clearer understanding of the genetic contribution to IS.

Main Methods:

  • A systematic literature search was performed across major scientific databases (PubMed, Embase, Web of Science, CNKI, WanFang).
  • Meta-analysis techniques were employed to pool odds ratios (ORs) and 95% confidence intervals (CIs) for the selected PPAR-γ polymorphisms.
  • Statistical analyses included heterogeneity testing, cumulative and sensitivity analyses, and publication bias assessment.

Main Results:

  • Sixteen studies comprising 3786 cases and 5343 controls were included.
  • The PPAR-γ rs1801282 C/G polymorphism was significantly associated with an increased risk of IS (GG vs. CC: OR=2.17, 95%CI=1.09-4.35; GG vs. CC+CG: OR=2.15, 95%CI=1.07-4.32).
  • No significant association was found between the PPAR-γ rs3856806 C/T polymorphism and IS risk.

Conclusions:

  • The PPAR-γ rs1801282 C/G polymorphism likely plays a role in the occurrence of ischemic stroke.
  • These findings highlight a potential genetic marker for IS risk, but require validation through additional prospective studies.
