This study identifies micro RNA-301a-5p (miR-301a-5p) as a key regulator in chicken spermatogenesis. It directly targets transforming growth factor-beta 2 (TGFβ2), influencing germ cell development and the TGF-β signaling pathway.
Area of Science:
- Reproductive Biology
- Molecular Biology
- Genetics
Background:
- Spermatogenesis is a complex process regulated by numerous factors.
- The role of microRNAs (miRNAs) in poultry spermatogenesis remains largely unexplored.
Purpose of the Study:
- To investigate key miRNAs and their target genes in chicken spermatogenesis.
- To elucidate the regulatory mechanisms of miRNAs in germline cell differentiation.
Main Methods:
- Next-generation sequencing for miRNA expression profiling in chicken germline cells (PGCs, SSCs, spermatogonia, sperm).
- Reverse transcription quantitative PCR (RT-qPCR), dual-luciferase assays, and miRNA salience analysis.
- Identification of miRNA-target gene interactions and pathway analysis.
Main Results:
- Identified several key miRNAs involved in spermatogenesis, with miR-301a-5p being prominent in PGCs, SSCs, and spermatogonia.
- Confirmed direct binding of miR-301a-5p to transforming growth factor-beta 2 (TGFβ2).
- Demonstrated miR-301a-5p's role in the transforming growth factor-beta (TGF-β) signaling pathway and germ cell development.
Conclusions:
- This study provides the first evidence of miR-301a-5p directly regulating TGFβ2 during spermatogenesis in chickens.
- The findings enhance understanding of miRNA-mediated regulation of germline cell differentiation and male fertility in poultry.
