Background:

  • The Signal Transducer and Activator of Transcription 6 (STAT6) pathway plays a role in lymphoma pathogenesis.
  • Immunohistochemical expression of STAT6 and its phosphorylated form (pSTAT6) in systemic lymphomas requires further investigation.

Purpose of the Study:

  • To investigate the immunohistochemical expression of both non-phosphorylated STAT6 and phosphorylated STAT6 (pSTAT6) in a cohort of systemic lymphomas.
  • To determine if STAT6 and pSTAT6 expression patterns differ among lymphoma types.

Main Methods:

  • Utilized immunohistochemistry to detect STAT6 (clone YE361) and pSTAT6 (clone Y641) in 60 lymphoma samples.
  • Analyzed the cellular localization (cytoplasmic vs. nuclear) and frequency of STAT6 and pSTAT6 expression.

Main Results:

  • STAT6 (clone YE361) showed predominantly cytoplasmic expression in 23.3% of cases, associated with diffuse large B cell and marginal zone B cell lymphomas.
  • pSTAT6 (clone Y641) exhibited mainly nuclear expression in 45% of cases, significantly associated with follicular lymphoma, Hodgkin lymphoma, and angioimmunoblastic T-cell lymphoma.
  • No significant association was found between STAT6/pSTAT6 expression and PD-L1 status, clinicopathological data, or patient prognosis.

Conclusions:

  • The immunohistochemical expression patterns of STAT6 and pSTAT6 are distinct and vary significantly between different lymphoma subtypes.
  • These findings highlight differential STAT6 pathway activation in lymphomagenesis and suggest potential subtype-specific roles.
