    Background:

    • C-reactive protein (CRP) is a key acute-phase protein primarily synthesized by the liver.
    • The role of peripheral blood cells in CRP production has been less understood.

    Purpose of the Study:

    • To investigate the synthesis and secretion of CRP by human peripheral mononuclear cells in culture.
    • To identify which specific mononuclear cell populations are responsible for CRP production.
    • To explore the regulatory mechanisms involved in CRP synthesis by these cells.

    Main Methods:

    • Human peripheral mononuclear cells were isolated and cultured.
    • Cells were stimulated with 10-O-tetradecanoyl-phorbol-13-acetate (TPA) in the presence of [3H]glutamine.
    • CRP synthesis was assessed via immunoprecipitation and SDS-polyacrylamide gel electrophoresis (SDS-PAGE) analysis of radiolabeled proteins.

    Main Results:

    • Evidence obtained confirmed the synthesis and secretion of CRP by cultured peripheral mononuclear cells.
    • Radioactivity was exclusively detected in the CRP band, indicating de novo synthesis.
    • T-cell preparations were identified as the main CRP-synthesizing cells, stimulated by a monocyte-derived factor.

    Conclusions:

    • Peripheral mononuclear cells, particularly T-cells, are capable of synthesizing and secreting CRP.
    • Monocyte-derived factors, potentially involving arachidonate cascade metabolites, regulate this T-cell CRP synthesis.
    • This finding expands our understanding of CRP production beyond the liver, implicating immune cells in its systemic regulation.
