    Summary

Researchers developed monoclonal antibodies (MAbs) to identify B cell antigens, revealing distinct expression patterns on resting and activated B cells and in various B cell leukemias and lymphomas.

    Area of Science:

    • Immunology
    • Cell Biology
    • Hematology

    Background:

    • Human B cells express various surface antigens.
    • Distinguishing B cell subsets and malignancies requires specific cell surface markers.

    Purpose of the Study:

    • To generate and characterize monoclonal antibodies (MAbs) against novel B cell antigens.
    • To define the expression profiles of these antigens on normal and malignant B cells.

    Main Methods:

    • Generation of a panel of MAbs targeting human B cell antigens.
    • Analysis of antigen expression on resting and activated B cells (in vivo and in vitro).
    • Immunophenotypic characterization of B cell tumors (leukemias and lymphomas) using MAbs.

    Main Results:

    • MAbs L26 and L27 showed pan-B cell specificity.
    • Antigens L22 and L30 were specific to resting B cells.
    • Antigens L4 and L29 were induced upon B cell activation.
    • Interleukin-2 receptor (IL-2R) was detected on activated B cells (MAb L10).
    • Specific antigen profiles were identified for common acute lymphatic leukemia (CALL), B cell chronic leukemia (B-CLL), hairy cell leukemia (HCL), and B cell lymphomas.

    Conclusions:

    • Novel MAbs provide tools to differentiate B cell subsets and activation states.
    • Antigen expression patterns correlate with B cell differentiation and malignancy.
    • Phenotypic analysis of B cell tumors aids in classification and understanding disease biology.
