    Summary

    Beige (bg/bg) mice exhibit cytotoxic T lymphocyte (CTL) cells with abnormal morphology and reduced killing ability during lymphocytic choriomeningitis virus (LCMV) infection. These mice compensate by increasing the number of CTLs, suggesting impaired granule formation affects lytic function.

    Background:

    • Cytotoxic T lymphocytes (CTLs) are crucial for viral clearance.
    • Beige (bg/bg) mice possess a genetic mutation affecting cellular granule formation.
    • The role of CTLs in bg/bg mice during viral infections is not fully understood.

    Purpose of the Study:

    • To investigate the morphology and function of CTLs in bg/bg mice during lymphocytic choriomeningitis virus (LCMV) infection.
    • To determine if the bg/bg mutation impacts CTL lytic activity and compensatory mechanisms.
    • To explore the relationship between granule formation and CTL function.

    • Isolation and analysis of spleen leukocytes from LCMV-infected bg/bg, bg/+, and +/+ mice.

    • bg/bg mice exhibit CTLs with impaired lytic capacity due to aberrant granule formation during LCMV infection.
    • A compensatory increase in CTL numbers occurs in bg/bg mice to manage viral replication.
    • These findings highlight the critical role of granule formation in CTL effector function and host antiviral defense.
