    Background:

    • The monoclonal antibody KJ23a identifies T cell receptors (TCRs) using the V beta segment V beta 17a.
    • T cells expressing V beta 17a receptors frequently interact with the MHC class II protein, IE.

    Purpose of the Study:

    • To investigate the fate of T cells expressing V beta 17a in mice expressing IE.
    • To determine the mechanism of self-MHC tolerance in normal animals.

    Main Methods:

    • Analysis of T cell populations in the peripheral and thymic compartments of mice.
    • Flow cytometry to identify and quantify T cells expressing specific V beta segments.

    Main Results:

    • T cells expressing V beta 17a are selectively eliminated from the peripheral T cell pool and mature thymocytes in mice expressing IE.
    • These T cells are present in normal numbers within the immature thymocyte population of IE-expressing mice.

    Conclusions:

    • Self-MHC tolerance in normal animals is primarily mediated by clonal elimination, not suppression.
    • Tolerance induction to self-MHC may occur during thymic selection of immature thymocytes into the mature pool.
