    Background:

    • Androgenic hormones are known to influence immune function through receptor-mediated pathways.
    • The presence and role of androgen receptors in the human thymus were previously uncharacterized.

    Purpose of the Study:

    • To investigate the presence and characteristics of high-affinity androgen receptors in the human thymus gland.
    • To explore the potential functional role of these receptors in thymocyte activity.

    Main Methods:

    • Analysis of cytosolic extracts from whole human thymus and derived thymocyte preparations.
    • Sucrose density gradient centrifugation to characterize receptor sedimentation.
    • Flow cytometry for assessing thymocyte purity.

    • Purified human thymocytes ( >99.3% pure) contained significant receptor levels (3.34 +/- 1.2 fmol/mg protein).
    • Mature lymphocytes and T cell lines (Jurkat) lacked detectable androgen receptors.
    • Dihydrotestosterone treatment reduced interleukin-2-like activity in cultured thymocytes.

    Conclusions:

    • This study provides the first evidence of a high-affinity androgen receptor in maturing human thymocytes.
    • The presence of these receptors suggests a functional role in thymocyte development and immune regulation.
    • Androgen modulation of interleukin-2 production indicates a potential mechanism for immune system influence.
