Treatment Resistant Cancers02:56

Treatment Resistant Cancers

3.4K
Cancer is the second leading cause of death in the United States. A cancer cell is genetically unstable and hence can mutate faster. They can also modify their microenvironment and escape immune surveillance. The difficulties in treating cancer are further compounded by the emergence of rapid resistance to anticancer drugs. The most common ways to attain resistance in cancer cells include alteration in drug transport and metabolism, modification of drug target, elevated DNA damage response, or...

Treatment for Pulmonary Arterial Hypertension: Endothelin Receptor Antagonists

238
Endothelins (ETs) are potent vasoactive peptides critical in the human body's various physiological and pathological processes. One of the most promising therapeutic strategies for treating pulmonary arterial hypertension (PAH) involves counteracting the effects of these endothelins using a class of drugs known as endothelin receptor antagonists.
ETs are synthesized through a complex sequence of enzymatic steps, primarily involving an enzyme referred to as endothelin-converting enzyme...

Combination Therapies and Personalized Medicine02:50

Combination Therapies and Personalized Medicine

5.2K
Combining two or more treatment methods increases the life span of cancer patients while reducing damage to vital organs or tissue from the overuse of a single treatment. Combination therapy also targets different cancer-inducing pathways, thus reducing the chances of developing resistance to treatment.
The combination of the drug acetazolamide and sulforaphane is a good example of combination therapy to treat cancer. The cells in the interior of a large tumor often die due to the hypoxic and...

Targeted Cancer Therapies02:57

Targeted Cancer Therapies

7.9K
The targeted cancer therapies, also known as “molecular targeted therapies,” take advantage of the molecular and genetic differences between the cancer cells and the normal cells. It needs a thorough understanding of the cancer cells to develop drugs that can target specific molecular aspects that drive the growth, progression, and spread of cancer cells without affecting the growth and survival of other normal cells in the body.
There are several types of targeted therapies against...

Tumor Immunotherapy01:27

Tumor Immunotherapy

712
Immunotherapy is a treatment that boosts or manipulates the immune system to fight diseases, including cancer. For instance, by stimulating an immune response through vaccinations against viruses that cause cancers, like hepatitis B virus and human papillomavirus, these diseases can be prevented. Nonetheless, some cancer cells can avoid the immune system due to their rapid mutation and division. The immune response to many cancers involves three phases: elimination, equilibrium, and escape.

Cancer Therapies02:49

Cancer Therapies

8.0K
Cancer therapies are various modes of treatment, such as surgery, radiation therapy, and chemotherapy that are administered to cancer patients.
However, cancer treatments can pose several challenges, as therapies used to kill cancer cells are generally also toxic to normal cells. Moreover, cancer cells mutate rapidly and can develop resistance to chemical agents or radiation therapy. Besides, all types of cancer cells may not respond to the same therapy. Some cancer cells respond to one...

Area of Science:

  • Oncology
  • Clinical Trials
  • Cancer Therapeutics

Background:

  • Treatment options for advanced endometrial cancer post-platinum-based chemotherapy are not well-defined.
  • There is a critical need for effective therapies to improve outcomes in this patient group.

Purpose of the Study:

  • To evaluate the efficacy and safety of lenvatinib plus pembrolizumab compared to chemotherapy in patients with advanced endometrial cancer.
  • To determine the impact of this combination on progression-free survival and overall survival.

Main Methods:

  • A phase 3 randomized trial assigned 827 patients with advanced endometrial cancer to lenvatinib plus pembrolizumab or physician's choice chemotherapy.
  • Primary endpoints included progression-free survival and overall survival, assessed in all patients and the mismatch repair-proficient (pMMR) subgroup.
  • Safety was evaluated based on the incidence of adverse events.

Main Results:

  • Lenvatinib plus pembrolizumab demonstrated significantly longer median progression-free survival (7.2 vs. 3.8 months) and overall survival (18.3 vs. 11.4 months) compared to chemotherapy.
  • These benefits were observed in both the overall patient population and the pMMR subgroup.
  • Grade 3 or higher adverse events were more frequent in the lenvatinib plus pembrolizumab arm (88.9%) versus chemotherapy (72.7%).

Conclusions:

  • Lenvatinib plus pembrolizumab significantly improves progression-free survival and overall survival in advanced endometrial cancer patients previously treated with platinum-based chemotherapy.
  • This combination therapy represents a promising new treatment option for this challenging patient population.
  • The study provides robust evidence supporting lenvatinib plus pembrolizumab as a new standard of care.
