Summary

Pomalidomide-based therapy shows promise for treating extramedullary disease (EMD) in multiple myeloma (MM) patients. This approach achieved significant response rates and improved survival, offering a new option for difficult-to-treat EMD.

Background:

  • Extramedullary disease (EMD) in multiple myeloma (MM) presents a significant challenge, often associated with poor prognosis.
  • Effective treatment strategies for EMD, particularly with novel agents, remain limited.
  • Pomalidomide is a novel immunomodulatory drug with potential in MM treatment.

Purpose of the Study:

  • To evaluate the efficacy of pomalidomide-based therapy in patients with multiple myeloma and extramedullary disease.
  • To assess response rates and survival outcomes in this patient population.

Main Methods:

  • Retrospective analysis of six patients with EMD treated with pomalidomide-based therapy.
  • Evaluation of serological and extramedullary response rates.
  • Assessment of progression-free survival (PFS) and overall survival (OS).

Main Results:

  • An overall response rate (ORR) of 83% was observed for extramedullary disease, with 50% complete response (CR) and 33% partial response (PR).
  • Serological response included 16% CR and 67% PR.
  • Median PFS was 5 months and median OS was 8 months from EMD diagnosis.

Conclusions:

  • Pomalidomide-based therapy demonstrated efficacy in previously treated patients with EMD.
  • These findings suggest pomalidomide as a viable treatment option for refractory EMD in MM.
