Summary

Serum cholesterol levels correlate with Ferroxidase II (Fox II) activity. This study found cholesterol potentiates Fox II development, distinct from caeruloplasmin (ferroxidase I).

Background:

  • Ferroxidase II (Fox II) is an enzyme found in serum.
  • Its relationship with serum lipids, particularly cholesterol, requires further elucidation.

Purpose of the Study:

  • To investigate the development of Ferroxidase II (Fox II) in serum.
  • To determine the correlation between Fox II activity and serum cholesterol concentration.
  • To examine the influence of cholesterol on Fox II activity in vitro.

Main Methods:

  • Serum samples were acid-incubated for 24 hours to develop Ferroxidase II (Fox II) activity.
  • Serum cholesterol concentrations were measured.
  • In vitro addition of cholesterol to serum was performed to assess its effect on Fox II.

Main Results:

  • A strong positive correlation was observed between developed Fox II activity and serum cholesterol levels in both normal subjects and hyperlipidaemic patients.
  • In vitro addition of cholesterol to serum confirmed its potentiating effect on Fox II development.
  • No significant correlation was found between developed Fox II and caeruloplasmin (ferroxidase I), nor between cholesterol and caeruloplasmin.

Conclusions:

  • Serum cholesterol concentration is a significant factor in the development of Ferroxidase II (Fox II) activity.
  • Cholesterol's potentiating effect on Fox II is independent of caeruloplasmin (ferroxidase I).
  • These findings suggest a specific role for cholesterol in modulating Fox II activity in serum.
