Progabide for refractory partial epilepsy: a controlled add-on trial.

D Schmidt, K Utech

    Summary

Progabide, an experimental antiepileptic drug, did not significantly reduce seizures in adults with chronic partial epilepsy. It increased levels of other drugs and had mild side effects, suggesting it is not ideal for treatment-resistant epilepsy.

    Area of Science:

    • Neurology
    • Pharmacology

    Background:

    • Epilepsy is a neurological disorder characterized by recurrent seizures.
    • Partial epilepsy is a common form, often requiring intensive treatment.
    • Developing effective treatments for drug-resistant epilepsy remains a challenge.

    Purpose of the Study:

    • To evaluate the efficacy and safety of progabide in adult patients with chronic partial epilepsy.
    • To assess progabide's impact on seizure frequency and its interaction with other antiepileptic drugs.

    Main Methods:

    • A placebo-controlled, double-blind, cross-over trial was conducted.
    • 19 adult patients with refractory chronic partial epilepsy participated.
    • Progabide was administered at a mean daily dose of 32 mg/kg.

    Related Experiment Videos

    Main Results:

    • Progabide did not significantly alter seizure frequency.
    • Therapeutic responses correlated with increased plasma concentrations of phenytoin and phenobarbital.
    • Carbamazepine comedication was linked to a poor response.
    • Mild side effects were observed, with one case of elevated SGOT/SGPT requiring drug withdrawal.

    Conclusions:

    • Progabide demonstrated no significant efficacy in reducing seizures for this patient group.
    • Potential drug interactions and specific side effects warrant careful consideration.
    • Progabide is unlikely to be a primary treatment option for patients with refractory epilepsy.
