Abnormal Proliferation02:23

Abnormal Proliferation

4.7K
Under normal conditions, most adult cells remain in a non-proliferative state unless stimulated by internal or external factors to replace lost cells. Abnormal cell proliferation is a condition in which the cell's growth exceeds and is uncoordinated with normal cells. In such situations, cell division persists in the same excessive manner even after cessation of the stimuli, leading to persistent tumors. The tumor arises from the damaged cells that replicate to pass the damage to the...

Cancer-Critical Genes II: Tumor Suppressor Genes

8.3K
Genes usually encode proteins necessary for the proper functioning of a healthy cell. Mutations can often cause changes to the gene expression pattern, thereby altering the phenotype.
When the function of certain critical genes, especially those involved in cell cycle regulation and cell growth signaling cascades, gets disrupted, it upsets the cell cycle progression. Such cells with unchecked cell cycles start proliferating uncontrollably and eventually develop into tumors.
Such genes that act...

The Retinoblastoma Gene01:20

The Retinoblastoma Gene

4.2K
Tumor suppressor genes are normal genes that can slow down cell division, repair DNA mistakes, or program the cells for apoptosis in case of irreparable damage. Hence, they play an essential role in preventing the proliferation of damaged cells.
The first-ever tumor suppressor gene called Rb was identified in retinoblastoma - a rare eye tumor in children. In inherited forms of the disease, a child inherits one defective copy of the Rb gene, which predisposes them to retinoblastoma. However,...

TGF - β Signaling Pathway01:16

TGF - β Signaling Pathway

7.9K
The TGF-β signaling pathway regulates cell growth, differentiation, adhesion, motility, and development. TGF-β ligands that induce TGF-β signaling are synthesized in their latent form. Several proteases or cell surface receptors such as integrins act upon the latent form, releasing the active ligand. There are three types of mammalian TGF-βs: (TGF-β1, TGF-β2, and TGF-β3) that bind as homodimers or heterodimers to TGF-β receptors. The TGF-β receptors...

Loss of Tumor Suppressor Gene Functions

5.2K
Tumor suppressor genes are normal genes that can slow down cell division, repair DNA mistakes, or program the cells for apoptosis in case of irreparable damage. Hence, they play an essential role in preventing the proliferation of damaged cells.
When the tumor suppressor genes develop mutations or are lost, cells start growing out of control, leading to cancer. However, a single functional copy of the tumor suppressor gene is enough for the cells to maintain their normal functions and cell...

Summary

Guanine nucleotide exchange factor T (GEFT) is abnormally expressed in most soft tissue sarcomas (STS). This aberrant GEFT expression, observed across various STS subtypes, may contribute to tumor development.

Area of Science:

  • Oncology
  • Molecular Biology
  • Genetics

Background:

  • Guanine nucleotide exchange factor T (GEFT) shows high amplification in rhabdomyosarcoma.
  • GEFT protein is overexpressed in rhabdomyosarcoma compared to normal striated muscle.
  • This study investigates GEFT expression in various soft tissue sarcomas (STS).

Purpose of the Study:

  • Evaluate aberrant GEFT expression in multiple STS subtypes.
  • Compare GEFT expression (protein and mRNA) between chromosomal translocation-associated sarcomas (CTAS) and non-chromosomal translocation-associated sarcomas (NCTAS).
  • Analyze differences in clinical pathology and histological features related to GEFT expression.

Main Methods:

  • Immunohistochemistry (IHC) and tissue microarrays for GEFT protein detection.
  • Quantitative reverse transcriptase polymerase chain reaction (qRT-PCR) for GEFT mRNA expression analysis.

Main Results:

  • GEFT positive expression (89.50%) and overexpression (51.60%) were significantly higher in STS than in normal striated muscle.
  • Aberrant GEFT expression was observed across all evaluated STS subtypes.
  • No significant difference in GEFT expression was found between CTAS and NCTAS.

Conclusions:

  • Abnormal GEFT expression is prevalent in diverse STS subtypes.
  • GEFT may play a crucial role in the tumorigenesis of soft tissue sarcomas.
