Background:

  • Ulcerative colitis (UC) is an inflammatory bowel disease often inadequately managed by traditional therapies.
  • Endometrial regenerative cells (ERCs) possess immunosuppressive properties.
  • Stem cell-derived conditioned media (CM) offer advantages over cell therapy, but ERC-CM efficacy for colitis is unexplored.

Purpose of the Study:

  • To investigate the therapeutic potential of ERC-CM in a mouse model of acute colitis.
  • To evaluate the immunomodulatory effects of ERC-CM on immune cells and cytokine profiles.

Main Methods:

  • Acute colitis was induced in mice using dextran sodium sulfate (DSS).
  • ERC-CM was administered to mice on days 4, 6, and 8 post-induction.
  • Disease activity, histological changes, immune cell populations (Tregs, M1 macrophages, Th1/Th17 cells), and cytokine levels (TNF-α, IL-6, iNOS, IL-10, IL-27) were assessed.
  • In vitro lymphocyte proliferation was measured.

Main Results:

  • ERC-CM treatment significantly ameliorated colitis symptoms and improved colon histology.
  • ERC-CM increased regulatory T cells (Tregs) while decreasing M1 macrophages and Th1/Th17 cells in the spleen and colon.
  • Local and systemic inflammatory markers (TNF-α, IL-6, iNOS, IL-17) were reduced, while anti-inflammatory cytokines (IL-10, IL-27) increased.
  • ERC-CM inhibited lymphocyte proliferation in vitro.

Conclusions:

  • ERC-CM demonstrates significant therapeutic effects in a colitis model, comparable to ERC therapy.
  • ERC-CM exhibits potent immunomodulatory and anti-inflammatory properties.
  • ERC-CM represents a promising cell-free therapeutic strategy for ulcerative colitis treatment.
