Ribosome Profiling02:24

Ribosome Profiling

3.7K
Ribosome profiling or ribo-sequencing is a deep sequencing technique that produces a snapshot of active translation in a cell. It selectively sequences the mRNAs protected by ribosomes to get an insight into a cell’s translation landscape at any given point in time.
Applications of ribosome profiling
Ribosome profiling has many applications, including in vivo monitoring of translation inside a particular organ or tissue type and quantifying new protein synthesis levels.
The technique...

Leaky Scanning02:28

Leaky Scanning

5.3K
During most eukaryotic translation processes, the small 40S ribosome subunit scans an mRNA from its 5' end until it encounters the first start AUG codon. The large 60S ribosomal subunit then joins the smaller one to initiate protein synthesis. The location of the translation initiation is largely determined by the nucleotides near the start codon as there may be multiple translation initiation sites present on the mRNA.  Marilyn Kozak discovered that the sequence RCCAUGG (where R...

Area of Science:

  • Genomics and Molecular Biology
  • RNA Biology
  • Bioinformatics

Background:

  • Small open reading frames (sORFs) are prevalent in various transcripts, often overlooked as non-functional.
  • Emerging evidence links sORFs to physiological functions and human diseases, suggesting therapeutic potential.
  • sORFs can exert function through encoded peptides or regulatory roles during translation.

Purpose of the Study:

  • To review high-throughput methods for sORF detection and characterization.
  • To discuss techniques for validating and functionally characterizing sORFs.
  • To emphasize the importance of including sORFs in gene annotations.

Main Methods:

  • Ribosome profiling to identify actively translated sORFs.
  • Mass spectrometry to detect sORF-encoded peptides.
  • Advanced ribosome profiling techniques to study translation initiation regulation by sORFs.

Main Results:

  • High-throughput techniques have streamlined sORF identification.
  • Defining the precise function of identified sORFs remains a significant challenge.
  • sORFs are found in diverse RNA types, including coding and non-coding RNAs.

Conclusions:

  • sORFs represent a substantial, underappreciated source of functional genetic elements.
  • Systematic characterization of sORFs is crucial for understanding their roles in health and disease.
  • Further research into sORF function may uncover novel therapeutic targets.
