    Summary

    The Kx antigen, typically found on red blood cells, is not present on human granulocytes. This study used multiple methods to confirm the absence of the Kx antigen on granulocytes, including those from patients with chronic granulomatous disease.

    Background:

    • The Kx antigen is a known red blood cell antigen.
    • Previous reports suggested the presence of Kx antigen on granulocytes, but this requires further investigation.

    Purpose of the Study:

    • To investigate the presence or absence of the Kx antigen on human granulocytes.
    • To clarify the relationship between the Kx antigen and granulocyte function, particularly in chronic granulomatous disease (CGD).

    Main Methods:

    • Utilized three techniques to measure anti-Kx uptake: 125I-staphylococcal protein A (125I-SPA), avidin-biotin-complex (ABC) immunoperoxidase staining, and granulocyte adsorption assays.
    • Adsorption of anti-Kx serum with purified granulocytes was performed to remove cross-reacting antibodies.
    • Tested granulocytes from normal donors, a Ko individual, and a patient with X-linked CGD.

    Main Results:

    • All three methods consistently indicated the absence of the Kx antigen on normal human granulocytes.
    • Initial reactivity was attributed to contamination of anti-Kx serum with an antibody to a granulocyte-specific antigen.
    • Adsorbed anti-Kx serum showed no change in reactivity with Kx-expressing red blood cells or McLeod red cells.
    • Granulocytes from normal donors, a Ko individual, and a CGD patient were non-reactive with adsorbed anti-Kx serum.

    Conclusions:

    • The Kx antigen is most likely a red cell-specific antigen.
    • The red cell Kx antigen is not directly related to the biochemical defect in X-linked chronic granulomatous disease (CGD).
    • These findings challenge previous reports suggesting Kx antigen presence on granulocytes.
