Summary
Aire deficiency impairs thymic regulatory T cell (Treg) production by causing medullary thymic epithelial cells (mTECs) to express CTLA-4. This disrupts T cell receptor signaling, leading to autoimmunity, but can be rescued by removing CTLA-4 from mTECs.
